We have to replace the main floor beam in a property we just purchased. 1 half of the building is about 35 feet, the second is about 16 feet. Currently there is what appears to be one beam running the length of the building.

We would like to install a wall in the basement to divide the two areas so would like to know if we can build 1 beam thats 16 feet long, and another thats 35 feet long and have them meet on a 2x8, 4 layer post thats built into the dividing wall? We plan to support both beams every 8 feet with jack posts but are not sure about having the 2 beams meet and how best to support them?

Yes IMO this would be fine to join the two beams over this 4 ply post.
